1962: When Hofmann Brought Synthetic Psilocybin Back to Maria Sabina
By Microdosing College Editorial·Last updated May 6, 2026·7 min read
The hofmann maria sabina 1962 synthetic psilocybin encounter is one of the most cited moments in psychedelic history — a scene in which laboratory chemistry and indigenous ritual met face to face, and a Mazatec healer quietly judged them equal. To understand why that moment mattered, it helps to trace the chain of events that made it possible.
For a broader account of how this period fits into the longer arc of mushroom use, see our history of psychedelic mushrooms pillar guide.
From Oaxaca to Sandoz and Back Again
The story begins in 1955, when banker and amateur ethnomycologist R. Gordon Wasson became one of the first Westerners to participate in a Mazatec mushroom ceremony — a — under the guidance of Maria Sabina in the mountain village of Huautla de Jiménez, Oaxaca, Mexico. Two years later, Wasson's account in magazine introduced millions of American readers to what he called "magic mushrooms" (for details on that publication, see our companion piece on ). Wasson brought mushroom specimens back to Paris, where mycologist Roger Heim cultivated them and passed samples to Albert Hofmann at Sandoz Laboratories in Basel.
Hofmann — already the discoverer of LSD in 1943 — accepted the challenge. Working with his Sandoz colleagues, he isolated the active compounds, identifying two related alkaloids. In 1958 he published the isolation and chemical characterization of psilocybin and psilocin, naming the compounds and establishing their structures. Sandoz subsequently manufactured synthetic psilocybin under the trade name Indocybin, making the compound available to researchers in precisely measured pill form. You can read more about that achievement in our article on Albert Hofmann's isolation of psilocybin in 1958.
The natural next question was whether the synthetic molecule, stripped of every other compound in the mushroom, would behave the same way — especially in the ritual context that gave the mushroom its meaning.
The 1962 Expedition to Huautla
In the summer of 1962, Hofmann and Wasson returned to Huautla de Jiménez together. They brought with them pharmaceutical-grade psilocybin pills synthesized at Sandoz — a controlled, measured quantity of the isolated compound with none of the organic material of the original mushroom. Their aim was straightforward: to offer Maria Sabina the pills in place of the mushrooms during a nighttime ceremony and observe what she reported.
Maria Sabina was by then in her sixties, a recognized curandera whose veladas had been conducted for healing purposes within the Mazatec tradition for decades. The velada is a nocturnal ceremony combining ingestion of the niños santos (the sacred mushrooms, teonanácatl), song, prayer, and divination; its structure and intent are inseparable from the mushrooms themselves. For a fuller account of that ceremony, see our explainer on the Mazatec velada ceremony.
According to accounts Wasson published and later recounted in several sources — including the collaborative book The Road to Eleusis (Wasson, Hofmann, and Ruck, 1978) — Maria Sabina consumed the Sandoz pills and conducted the ceremony. Her reported verdict, as relayed through translators, was that the spirit of the mushroom was present in the pills. She is said to have affirmed that the pills "worked" in the way the mushrooms worked, that the deity or spirit she encountered was the same.
This was precisely what Hofmann needed to hear as a pharmacologist. If the Mazatec healer, whose entire relationship with the mushrooms was embedded in spiritual and medical tradition rather than chemistry, reported the same essential experience from isolated psilocybin as from the whole mushroom, it was strong — if informal — evidence that psilocybin was indeed the primary active compound responsible for the effects the mushrooms had produced for generations.
What the Verdict Actually Established
It is worth being careful about what the 1962 encounter did and did not prove. Maria Sabina was not running a blinded pharmacological trial. She knew she was taking something Hofmann had brought; she was told it came from the mushroom. The conditions were ceremonial, not controlled. Her statement, filtered through translation and reported at second hand in Wasson's published accounts, cannot bear the full evidential weight sometimes placed on it.
What it did offer was corroborating testimony from a deeply experienced practitioner that the qualitative character of the experience was continuous with the mushroom experience — not wildly different, not inferior, not alien. Combined with Hofmann's chemical characterization and the clinical data Sandoz was beginning to gather with Indocybin in psychiatric research settings, it made a reasonable early case that psilocybin was not merely one of several equally important compounds but the central active agent.
The encounter also raised questions that pharmacology has revisited ever since. The mushroom contains not only psilocybin but psilocin, baeocystin, norbaeocystin, and trace amounts of other compounds. The theory that these may interact to produce an effect different from pure psilocybin alone — sometimes called the entourage effect — is still discussed, though evidence for it in the psychedelic context remains limited and contested. Maria Sabina's reported equivalence verdict is sometimes cited by those skeptical of the entourage hypothesis, and sometimes dismissed as culturally and methodologically insufficient to settle the question.
The Human Cost and Maria Sabina's Later Years
The 1962 visit was not the last time outsiders came to Huautla. Wasson's 1957 Life article had already set off a slow trickle of seekers; through the 1960s that trickle became a flood. Beatniks, hippies, and researchers arrived in numbers the village had never seen. Local authorities held Maria Sabina partly responsible for the disruption. Her home was reportedly burned. She was, in the words of various ethnographic accounts, blamed by her own community for opening a sacred practice to the outside world.
Maria Sabina herself, in statements recorded by the Mexican poet Álvaro Estrada for his 1977 biography María Sabina: Su vida y sus cantos (published in English as Maria Sabina: Selections), expressed ambivalence. She is quoted as saying the mushrooms had lost their power because they had been profaned — used by people without sickness to heal, without knowledge to guide, for curiosity rather than need. Whether that judgment extended to Hofmann's pills specifically is unclear; her reported 1962 approval and her later lamentations both belong to the record, and they need not contradict each other.
Wasson, for his part, later expressed regret about the consequences of the Life article for Huautla and for Maria Sabina personally, though he continued to write about and advocate for mushroom research until his death in 1986.
A Footnote That Still Echoes
The 1962 encounter between Hofmann, Wasson, and Maria Sabina occupies an unusual place in the history of psychedelic pharmacology. It is not a clinical study and was never presented as one. But as a moment of cross-cultural pharmacological inquiry — a synthesis of Mazatec ceremonial knowledge and Swiss laboratory chemistry — it has few equivalents.
Hofmann's own account of the journey appears in his 1979 book LSD: My Problem Child (German original LSD — mein Sorgenkind), in which he describes the expedition with evident reverence for both the tradition and the molecule. Sandoz's synthetic psilocybin — produced in the same Basel laboratory where Hofmann had isolated the compound in 1958 — had, in his telling, passed the most important test he could imagine: it had been recognized by the woman who embodied the tradition from which the compound came.
For anyone tracing the lineage from Mazatec velada to modern psilocybin clinical trials, that recognition matters. The history of psychedelic mushrooms is rarely a straight line, but the 1962 meeting in Huautla is one of its cleaner junctions — the point where indigenous knowledge, mid-century pharmacology, and the question of what makes a medicine "the same" all briefly converged.
Frequently asked
What happened when Albert Hofmann brought synthetic psilocybin to Maria Sabina in 1962?+
In the summer of 1962, Hofmann and Wasson traveled to Huautla de Jiménez and offered Maria Sabina pharmaceutical-grade psilocybin pills — synthesized by Sandoz — during a nighttime velada ceremony. According to accounts Wasson later published, she reported that the spirit of the mushroom was present in the pills, suggesting the synthetic compound produced a comparable experience to the whole mushroom.
Why was Maria Sabina's reported verdict pharmacologically significant?+
Her testimony offered early corroborating evidence that isolated psilocybin — stripped of all other mushroom compounds — was the primary agent responsible for the effects of the sacred mushrooms. It supported Hofmann's 1958 chemical characterization and informed Sandoz's clinical research with synthetic Indocybin pills throughout the 1960s.
Did the 1962 encounter prove that synthetic psilocybin is identical in effect to whole mushrooms?+
No. The 1962 encounter was not a controlled clinical trial. Maria Sabina was not blinded to what she was taking, and her statements were filtered through translation and reported secondhand. It provided qualitative testimony from an experienced practitioner, not scientific proof. The question of whether whole-mushroom compounds interact to produce effects different from pure psilocybin — sometimes called the entourage effect — remains a live topic in psychedelic research.
What were the broader consequences for Maria Sabina and Huautla de Jiménez?+
Wasson's 1957 Life magazine article had already begun attracting outsiders to Huautla; through the 1960s that became a large influx. Maria Sabina reportedly faced community backlash, and her home was burned. In accounts recorded by Álvaro Estrada for his 1977 biography, she expressed that the mushrooms had lost their power because outsiders had used them without the knowledge or need the tradition required.
Where can I read Hofmann's own account of the 1962 expedition?+
Albert Hofmann describes the journey to Huautla in his 1979 book LSD: My Problem Child (German title LSD — mein Sorgenkind), which remains in print. Wasson's perspective appears across several of his later publications, including the collaborative volume The Road to Eleusis (1978), co-authored with Hofmann and Carl Ruck.
